LETHAL DOSE OF PARACETAMOL (ACETAMINOPHEN) DAMAGES BLOOD AND SPLEEN IN BROILER CHICKS

Abstract

Aim of this study was to find out toxic effects of paracetamol on extrahepatic tissues i.e the blood and spleen in 14 days old broiler chicks after administration of lethal dose at 24 hr. Chicks of control group received only vehicle (Gr I) while chicks of second group (Gr II) received paracetamol at lethal dose (2 gm/kg/bw, single intra muscular injection). After 24 hours blood was used for hematological investigation. Spleen was used for morphology and histology. Blood smear were made to find out micronuclei. Dark color of blood of treated chicks suggests possibility of methamoglobinemia. All haematological parameters studied were altered i.e values of haemoglobin, total RBC and MCHC declined but PCV, MCV, MCH and RDW increased. Poor staining and disappearance of RBC nucleus suggests direct damage to them. No micronuclei formation seen. Splenomegaly associated with its disorganization could be noticed. Results suggest that paracetamol at lethal dose also damages extra hepatorenal tissues like blood and spleen. Probable role of oxidative stress caused by paracetamol and possible involvement of mitochondria in RBC is discussed.
