To investigate the effects of D-002 (beeswax alcohols) on esophagitis induced by duodenoesophageal reflux (DER) and duodenogastroesophageal reflux (DGER) in rats. In DER rats were randomized into six groups: a negative control and five with DER: a positive control, three D-002- (50, 200 and 400 mg/kg, respectively) and one omeprazole (20 mg/kg) groups, all treated orally for 14 days after inducing DER. In DGER, rats were randomized into eight groups: a negative control and seven with DGER: a positive control, three treated with D-002 (200, 400 and 800 mg/kg, respectively) and one with omeprazole 20 mg/kg, all given for 10 days before inducing DGER and during the next 48 h after the procedure. Esophageal lesions’ index (ELI) and macroscopic changes (severe inflammation) were determined. Positive controls with DER-induced esophagitis exhibited increased ELI values and macroscopical lesions (esophagitis and severe inflammation) as compared to negative controls. D-002 (50, 200 and 400 mg/kg) lowered significantly ELI values (45.8, 54.7 and 59. 2%, respectively) and inflammation (87.5, 77.8 and 77.8 % respectively). Effects on ELI, not on inflammation, were dose-dependent. Reductions of esophagitis changes, however, were not significant. D-002 (200 - 800 mg/kg) failed to ameliorate the changes induced by DGER. Omeprazole (20 mg/kg) was effective to reduce ELI, esophagitis and inflammation induced by DER and DGER. Repeat oral doses of D-002 (200 – 800 mg/kg) ameliorated DER-induced esophagitis in rats, while failed to reduce DGERinduced esophagitis.
