Among the oral anti-hyperglycemic agents (anti-diabetic drug) dipeptidyl peptidase-4 (DPP-4) inhibitor is an emerging class of drugs with vildagliptin in its forefront. This study aims to evaluate the Antioxidant activity of Vildagliptin using DPPH assay. DPPH radical scavenging activity was done using the method of Yohozowa et al. The reaction mixture containing 1.9ml of DPPH solution (150µM in ethanol) with different concentrations of the substance (500, 400, 200, 100, 50, µg/0.1ml) was shaken and incubated in dark for 30min at room temperature. The resultant absorbance was recorded at 517nm. At concentrations of 50, 100, 200, 300, 400 and 500 µg/0.1ml the percentage of inhibition observed was 58.37±0.67%, 63.30±1.23%, 68.84±0.85%, 68.63±0.60%, 69.84 ±0.37% and 70.54±0.52 % respectively. Our study shows that Vildagliptin has significant free radical scavenging activity which can contribute to its wide range of therapeutic profile.
